48份新疆野生狗牙根遗传多样性的SRAP分析

【摘要】 采用SRAP标记对48份新疆野生狗牙根材料进行遗传多样性分析,14对SRAP引物共扩增产生了133条带,其中123条显示多态性,因此SRAP标记可以有效地用于狗牙根野生资源遗传多样性评价。在物种水平上,多态位点百分率为77.83%,48份狗牙根材料的GS值为0.276~0.930,平均为0.700,各材料间差异明显,具有较为丰富的遗传多样性;居群间遗传分化系数为0.391 4,表明有39.14%的变异存在于居群间,60.86%的变异存在于居群内,居群内的遗传分化大于居群间的遗传分化;聚类分析表明,48份供试材料相似系数为0.58时可被聚为4类,聚类结果与地理来源有一定的相关性,但也有材料例外。

【Abstract】 The genetic diversity of Cynodon dactylon L.Pers from Xinjiang was investigated by using SRAP marker,133 bands were amplified by 14 pairs of informative and reliable primers,of which 123 were polymorphic.So,SRAP marker can be used to estimate genetic diversity of Cynodon dactylon L.effectively.At species level,percentage of polymorphic sites is 77.83%;GS value of 48 Cynodon dactylon materials were 0.276-0.930,whose average was 0.700.There were significant differences among all the materiats,with richer genetic diversity.The genetic differentiation coefficient in populations were 0.391 4,which showed there were variances of 79.14% in populations,60.86% genetic variation existed within populations.This data illustrates genetic variation within population is larger than that among populations.Genetic similarity of 48 materials ranges from 0.276 to 0.930.The variation among all materials are obvious,which shows they have rich genetic diversity.The general clustering consequences indicated 48 materials planted in Xinjiang were classified into four groups when the similarity coefficient is 0.58,the results is correlated with geography source,but some materials are exceptional.
