【摘要】 目的:研究骨桥蛋白(OPN)在硬皮病患者局部皮损表达和分布情况,探讨其在硬皮病中发挥的作用。方法:采用免疫组化SABC法分别检测硬皮病皮损20例(系统性硬皮病12例,局限性硬皮病8例)和正常对照组皮肤10例中OPN蛋白的表达情况。结果:OPN蛋白在硬皮病患者皮损真皮层的成纤维细胞中表达明显增高,20例硬皮病患者OPN阳性率为80%,而正常对照组仅有2例基底层呈微弱表达,阳性率为20%,两组阳性率比较有显著性差异(P<0.01);但16例阳性表达中系统性硬皮病10例,局限性硬皮病6例,OPN在系统性硬皮病患者和局限性硬皮病患者之间表达无明显差异(P>0.05)。结论:OPN蛋白在硬皮病患者皮损内明显高表达,并定位与成纤维细胞提示OPN可能参与了硬皮病的发病机制,对纤维化的形成发挥一定的作用,并对局限性和系统性硬皮病有相似的作用机制。更多还原

【Abstract】 Objective:To explore the expression and effect of local lesions of scleroderma patients. Methods:The expression of osteopontin was examined by immunohistochemical analysis in serial sections of scleroderma biopsies obtained from 20 patients (12 of systemic scleroderma ,8 of local scleroderma) ,and 10 normal controls.Results:Significant increase of osteopontin expression in fibroblasts was found in skin lesions of patients with scleroderma compared to normal controls (p<0.01). However osteopontin expression in systemic scleroderma showed no significance difference compared to local scleroderma (p>0.05).Conclusions:High expression of OPN in skin lesions of patients with scleroderma suggest that OPN might take part in the pathogenesis and have an important role in the fibrosis of scleroderma.更多还原
