黄芪种子和种苗质量标准研究

【摘要】 黄芪为常用大宗药材之一,入药的基原植物为蒙古黄芪(Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao))和膜荚黄芪(Astragalus membranaceus(Fisch.)Bge.)。本文对蒙古黄芪和膜荚黄芪种子检验方法进行了试验研究,确定了种子质量检验方法;通过对主要产区种子样品的净度、发芽率、含水量和其他植物种子含量等标进行检验和分析,对黄芪种子质量分级制定了标准;对蒙古黄芪种苗样品的根粗、根长和越冬芽长度进行了测定分析,并对蒙古黄芪种苗质量分级制定了标准。论文获得以下结果:1、蒙古黄芪和膜荚黄芪种子重量均采用千粒重法测定,最小种子净度检测量为25g,最小送检样品重量为250g。种子生活力最适检验方法为TTC染色法,TTC溶液适宜浓度为0.5%,在30℃的条件下染色120min;在最适发芽温度为14~20℃变温下的纸上发芽,初次计数时间为第3天,末次计数时间为第11天;水分测定采用高温(130±2℃)法烘干3h。2、依据黄芪种子的净度、发芽率、含水量和其他植物种子数4项指标将种子质量划分为3级,质量标准为1级种子净度≥98%,发芽率≥95%,含水量≤8%,其他植物种子数≤500粒/kg;2级种子净度≥95%,发芽率≥90%,含水量≤8%,其他植物种子数≤1000粒/kg;3级种子净度≥90%,发芽率≥80%,含水量≤8%,其他植物种子数≤1500粒/kg。3、蒙古黄芪种苗质量根据根长、根直径和芽长3项指标将其分为4个等级,1级种苗直径≥0.8cm,苗长≥30cm,芽长≤1cm;2级种苗直径≥0.6cm,苗长≥25cm,芽长≤1.5cm;3级苗直径≥0.4cm,苗长≥20cm,芽长≤2cm;4级苗直径≥0.4cm,苗长18~20cm,芽长≤2.5cm。

【Abstract】 Astragalus is a bulk medicinal material commonly used,and the basic plant used for it is Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)Hsiao and Astragalus membranaceus(Fisch.)Bge.This paper will illustrate the experiment and conclusion followed for testing methods of the seeds of Astragalus membranaceus(Fisch.)Bge.var.mongholicus(Bge.)and Astragalus membranaceus(Fisch.)Bge.Through testing and analyzing the purity,germination rate,water content,and other plant seed content of the seed samples from major producing regions,the seed quality classification standards for Astragalus membranaceus were established.Through measuring and analyzing the root and root lengths and the lengths of overwintering buds of A.var.mongholicus seedlings,the quality grading standards for A.var.mongholicus seedlings were formulated.The paper obtained the following results:1.The quality testing of seeds of A.var.mongholicus and Astragalus membranaceus can be implemented in the same way,including index as from germination rate,seed viability,water content and 1000-seed weight.The two kinds of seed weight methods assume to be 1000-grain method,with grain weight being of 5.102~7.009.Seed vigor optimum detection method should be TTC staining,the optimum concentration is 0.5% TTC solution in and the dyeing at 30 ℃being of 120 min.Seeds germinate the suitable condition is 15/25 temperature in paper bed.T℃ he initial count time was 3d,and the last count time was 10 d.The moisture content was measured by high temperature(130±2°C)drying method for 3h.2.Measured by index of clarity basis,Astragalus seed germination rate,moisture content and other plant seeds number,seed quality will be categorized as three levels: the first level is as grading standards for clarity a seed ≥98%,the germination rate ≥95%,water content ≤8%,seed number of other plants ≤500 seeds/Kg;the second is as seed ≥95%,germination rate ≥90%,moisture content ≤8%,number of other plant seeds ≤1000 seeds/Kg;the third grade is as seed clarity ≥ 90%,germination rate ≥ 80%,water content ≤ 8%,number of other plant seeds ≤ 1500/Kg.3.According to the 3 index of root length,root diameter and shoot length,quality of seedlings of A.var.mongholicus is divided into 4 grades: the primary seedling diameter is ≥0.8cm,with the seedling length being ≥30cm,and the shoot length ≤1cm;second seedlings’ diameter is ≥ 0.6cm,with seedling length ≥ 25 cm,and bud length ≤ 1.5cm;tertiary seedling diameter is ≥ 0.4cm,with seedling length being ≥ 20 cm,and bud length ≤ 2cm;the fourth seedling diameter is ≥ 0.4cm,with seedling length being 18~ 20 cm,and bud length ≤2.5cm.
