福寿螺消化系统的形态及菌群结构研究

【摘要】 目的:福寿螺对我国的农业生产造成极大的损害,是我国首批外来入侵物种之一但目前对福寿螺的基础研究较少。本试验研究其消化系统的形态结构和菌群结构,为福寿螺的深入研究奠定基础。方法:本实验通过对福寿螺消化系统的解剖及制作消化系统主要器官的组织切片来研究消化系统的形态结构;再通过变性梯度凝胶电泳(DGGE)分析雌、雄福寿螺胃和肠道的菌群结构,同时对优势条带进行回收、克隆及测序;进一步分离培养肠道中的细菌,并对细菌的生理生化特性及16SrDNA序列进行鉴定。结果:1福寿螺消化系统的形态结构:福寿螺的消化道非常发达,消化腺中有大量的分泌细胞,分泌活动旺盛,消化道有着发达的肌层且胃中存在软骨质的胃檐。2福寿螺消化系统的菌群结构:DGGE结果表明,雌、雄福寿螺胃和肠道中的优势菌几乎相同,优势条带的回收测序结果表明这些细菌分别属于气单胞菌属、希瓦氏菌属、粘质沙雷菌、肠杆菌属、虹鳟鱼肠道细菌及不可培养的细菌和不可培养的细菌克隆。3肠道细菌的分离培养:经过分离培养,得到两个菌株,结合细菌的生理生化鉴定和16SrDNA序列分析,结果表明分离到的细菌为产气肠杆菌和腐败希瓦氏菌。结论:首次对福寿螺消化系统的菌群结构进行研究,得出雌、雄福寿螺胃和肠道中的优势菌几乎相同,进一步分离培养了肠道中的细菌,为福寿螺消化系统的的研究提供了一定的基础。

【Abstract】 Objective:Pomacea canaliculata, which was included in the first batch of China’s invasion of alien species, cause great damage on China’s agricultural production. But less studies on the basis of Pomacea canaliculata,so in our study, studied the morphology and the bacterial population structure of the digestive system of Pomacea canaliculatas, and provide a theoretical basis for the study of Pomacea canaliculata.Methods:Studied the morphology of the digestive system of Pomacea canaliculatas by anatomy the digestive system and making paraffin section.And the bacterial population structure and diversity of gastrointestinal microbiota associated with the male and female Pomacea canaliculatas were analyzed by denaturing gradient gel electrophoresis (DGGE), and the main bands were recovered, cloned and sequencing. Intestinal bacteria from the Pomacea canaliculatas were isolated and cultured, identification of bacteria through physiological and biochemical identification and16SrDNA sequence identification.Results:1The morphology of the digestive system of Pomacea canaliculatas:There are a large number of secretory cells in the digestive gland of Pomacea canaliculatas, and Secretion exuberant. Pomacea canaliculatas’digestive tract has developed relatively thick muscular layer and there is cartilaginous gastric shield in the stomach, this is the basis of rapid physical and chemical digestion of Pomacea canaliculata.2The bacterial population structure of the digestive system of Pomacea canaliculatas:Flora bacteria is in much the same way between in stomach and in intestine of male and female Pomacea canaliculata. Sequencing results show these bacteria belonged to the genus Aeromonas, Shewanella, Serratia marcescens, Enterobacte^Rainbow trout intestinal bacterium, uncultured bacteria isolated by DGGE, non-culturable bacterial clones.3Isolation and culture of intestinal bacteria:Two strains of intestinal bacteria from the Pomacea canaliculatas were isolated and cultured, through physiological and biochemical identification and16SrDNA sequence identification, the results indicate that these two bacteria were Enterobacter aerogenes and corruption Shewanella.Conclusion:The bacterial population structure of the digestive system of Pomacea canaliculatas was first analyzed, and proof that the flora bacteria is in much the same way between in stomach and in intestine of male and female Pomacea, and further isolated and cultured the bacteria in the gut, the above research work provide a theoretical basis for Pomacea canaliculatas.
