电针对肥胖大鼠骨骼肌AMPK的影响
Effects of Electro-Acupuncture on Ampk in Skeletal Muscle of Obese Rats

【摘要】 目的:观察电针“后三里”和“内庭”对食源性肥胖大鼠的减肥效应,对骨骼肌AMPKα、 ACC、CPT-1表达的影响,分析探讨针灸减肥的作用机制。方法:将45只刚断乳的SD雄性大鼠随机分为两组:正常对照组15只,以普通饲料喂养;高脂组30只,以高脂饲料喂养。高脂致肥饲料喂养3个月后,以体重超过普通饲料组20%为肥胖标准,确定为食源性肥胖模型大鼠。将造模成功肥胖大鼠随机分为电针组和模型组,每组7只。正常对照组随机挑选7只。电针组:针刺一侧“后三里”和“内庭”,连接电针仪,频率2/15Hz,强度1mA,时间15min,左右侧隔日交替轮换取穴。模型组:与电针组同样束缚,但不电针;正常组:不做任何处理。每周治疗6天休息1天,共治疗4周。治疗期间每组大鼠均以普通饲料喂养。比较各实验组治疗前后大鼠体重、体脂、甘油三酯、总胆固醇、低密度脂蛋白的差异,运用实时定量PCR检测各组大鼠骨骼肌AMPKα mRNA、 ACCmRNA、CPT-1mRNA的表达情况,运用Western Blot检测各组大鼠骨骼肌AMPKα、 ACC、CPT-1蛋白活性。结果:电针对肥胖大鼠体重改善明显:治疗后电针组大鼠体重与模型组相比差异具有统计学意义(P<0.05),与正常组比较差异无显著性(P>0.05);治疗后电针组大鼠体重差与模型组相比差异有统计学意义(P<0.05),与正常组相比有显著性差异(P<0.01);治疗后电针组大鼠体重降低率与模型组、正常组相比存在显著性差异(P<0.01,P<0.01)。第1周治疗后,电针组大鼠体重与模型组比较差异有统计学意义(P<0.05),与正常组比较有显著性差异(P<0.01);但经第2、3周治疗后,电针组大鼠体重与模型组相比差异无统计学意义(P>0.05),第4周治疗后,电针组大鼠体重与模型组相比存在显著性差异(P<0.01)。电针对肥胖大鼠体脂改善明显:治疗后电针组大鼠内脏脂肪量与模型组相比有显著性差异(P<0.01),与正常组相比差异无显著性(P>0.05);电针组脂体比与模型组比较差异有统计学意义(P<0.05),与正常组相比差异无显著性(P>0.05);治疗后电针组大鼠项后棕色脂肪量与模型组相比差异有统计学意义(P<0.05),与正常组比较差异无显著性(P>0.05);电针组项后脂体比与模型组比较差异具有统计学意义(P<0.05),与正常组相比差异无显著性(P>0.05)。电针对肥胖大鼠血脂改善明显:治疗后电针组大鼠血清总胆固醇与模型组比较差异无统计学意义(P>0.05),正常组比较差异无统计学意义(P>0.05);电针组血清甘油三酯与模型组组比较差异有统计学意义(P<0.05),正常组比较差异有统计学意义(P<0.05);电针组低密度脂蛋白与模型组相比差异有统计学意义(P<0.05),与正常组比较有显著性差异(P<0.01)。电针对肥胖大鼠AMPKα-mRNA、ACC-mRNA、CPT-1-mRNA表达改善明显:肥胖大鼠AMPKα-mRNA、ACC-mRNA、CPT-1-mRNA的表达量与正常组比较均降低,治疗后电针组大鼠骨骼肌AMPKα-mRNA、ACC-mRNA、CPT-1-mRNA的表达量与模型组相比均提高。电针对肥胖大鼠骨骼肌AMPKα、PAMPKα、ACC、PACC、CPT-1表达改善明显:肥胖大鼠PAMPKα/AMPKα、PACC/ACC、CPT-1/β Actin值与正常组相比均降低,治疗后电针组PAMPKα/AMPKα、PACC/ACC、CPT-1/β Actin值与模型组相比均明显提高。结论:电针“后三里”和“内庭”对食源性肥胖大鼠有良好的减肥效应,降低肥胖大鼠升高的体脂和血脂,它能提高肥胖大鼠下降的骨骼肌AMPKα-mRNA、ACC-mRNA、CPT-1-mRNA的表达量,提高肥胖大鼠降低的P AMPKα/AMPKα、PACC/ACC、CPT-1/β Actin值,抑制脂肪合成,促进脂肪分解。
【Abstract】 Objective:To observe the effects of electro-acupuncture(EA) at ST36and ST45treating foodborne obese rats and influence on the expression of skeletal muscle Amp-activated protein kinase,Acetly-CoA carboxylase and carnitine palmitoyl transferase-1.Method:fat group30to high-fat diet.After the high-fat diet,induced fat three months,with10%of body weight than ordinary standard diet group were obese,defined as foodborne obese rats.The successful modeling obese rats were randomly divided into EA group and model group,each group has seven.Normal control group rats were randomly selected seven.EA group:acupuncture ST36and ST45, electro-acupuncture device is connected, the frequency2/15Hz, the intensity of1mA, time15min, alternating left and right side every other day rotation acupoints.Model group: Like bondage with electro-acupuncture group, but not electro-acupuncture;Normal group:without any treating.Treatment six days a week to rest one day, a total of4weeks of treatment.During treatment for each group of rats are fed with normal diet.Comparing rats’ body’s weight,body fat,TC,TQLDL-C.Expression of skeletal muscle AMPKa-mRNA, ACC-mRNA,CPT-1-mRNA were mersured by real-time quantity PCR.Result:Comparing electro-acupuncture groups with model group, the differences of each index have notable statistical significance (p<0.01), and comparing with normal group, there is no statistical significance (p>0.05). Between three electro-acupuncture groups, there exist differences, which is not so distinguished.Conclusion:electro-acupuncture at ST36and ST45can reduce body’s weight,body fat,blood lipids. increase the expression of P AMPKα/AMPKα、P ACC/ACC、CPT-1/βActin in skeletal muscle.Inhibiting the synthesis of fat and promote fat decomposition.
