西青果质量标准的建立

【摘要】 目的:建立、完善西青果的质量标准。方法:采用横切面显微法、薄层色谱法进行药材鉴别,采用高效液相法(HPLC)测定没食子酸的含量,色谱柱为Ultimate■XB-C18(GD)柱(250 mmx4.6 mm,5μm),流动相为甲醇-0.1%磷酸水溶液(5∶95),流速:1.0 ml·min-1,柱温:30℃,检测波长:273 nm。结果:西青果横切面具有索状组织、管状细胞、草酸钙簇晶等特征结构;薄层色谱表征化学成分的差异,两者均可用于西青果的定性鉴别;没食子酸在进样量为0.027~1.098μg范围内线性关系良好(r=1.000 0),平均加样回收率为97.02%,RSD为1.05%(n=6)。结论:本文基于薄层色谱鉴别、显微横切面鉴别、主成分没食子酸含量测定等系统优化检测方法可为进一步完善西青果质量标准提供依据。

【Abstract】 Objective: To establish and optimize the quality standard for Fructus Chebulae Immaturus. Methods: Microscopical characteristics were studied based on transverse section. TLC was adopted to identify Chebulae Fructus Immaturus. The content of gallic acid was determined by HPLC. A Welch Ultimate ■ XB-C18( GD)( 250 mm × 4. 6 mm,5 μm) column was used as the analytical column,the mobile phase was methanol-0. 1% phosphoric acid( 5 ∶ 95) at the flow rate of 1. 0 ml·min-1,the column temperature was 30℃ and the detection wavelength was 273 nm. Results: The funicular organization,tubular cells and calcium oxalate crystal cluster structure were found in the observation by the microscopic method. TLC characterization of differences could be used to identify the chemical compositions. Both of the methods mentioned above could be used for the qualitative identification of Chebulae fructus immaturus. The linear range of gallic acid was 0. 027-1. 098 μg( r = 1. 000 0),the average recovery was 97. 02%,and the RSD was 1. 05%( n = 6). Conclusion: The study optimized the TLC identification and established the microscopic transverse section identification and content determination method,which provide evidence for the further improvement of quality standard of Chebulae Fructus Immaturus.
