【摘要】 目的:探讨生姜泻心汤对伊立替康(CPT-11)所致大鼠迟发性腹泻的预防作用。方法:尾静脉注射CPT-11150mg/kg/d,连续2d,复制CPT-11所致大鼠迟发性腹泻模型,观察腹泻、体重及进食量的动态变化。健康SD大鼠18只随机分为正常对照组、腹泻模型组、中药+腹泻模型组。腹泻模型造模方法同上,正常对照组尾静脉注射等体积生理盐水。中药+腹泻模型组于注射CPT-11前3d开始,每天1次用生姜泻心汤灌胃,共用药9d。对各组大鼠腹泻情况进行评分,记录体重及进食量;注射CPT-11后d7将大鼠处死,取回肠、盲肠及结肠组织,光镜下观察肠粘膜组织结构。结果:大鼠尾静脉注射CPT-11后,出现明显的腹泻,并呈现早发性和迟发性腹泻两个时相阶段。中药治疗组迟发性腹泻评分(1.83±0.72)较模型组(2.19±0.94)显著下降(P<0.05),腹泻发生率也较模型组低,体重下降及进食量减少与模型组相比均得到显著改善(P<0.05,P<0.01)。光镜下肠粘膜损坏程度较模型组轻微。结论:生姜泻心汤对CPT-11诱发迟发性腹泻大鼠具有预防作用,并且可改善肠粘膜损伤程度。更多还原

【Abstract】 Objective:To study the preventive function of Zingiberis Recentis Decoction for Purging Stomach-Fire on delayed-onset diarrhea caused by irinotecan(CPT-11) in rats.Methods:Sprague Dawley male rats(n=6) injected with 150 mg/kg body weight/day CPT-11 for 2 consecutive days into the tail vein,to establish a model of delayed-onset diarrhea.Eighteen Sprague Dawley male rats were randomly assigned to three groups,normal group,delayed-onset diarrhea group and herb group.Herb group were treated with Zingiberis Recentis Decoction three days before CPT-11 injected and kept for 9 days.Scoring of delayed-onset diarrhea.Their body weight and food-intake were recorded.Rats were killed on day 7 after CPT-11 injection,intestinal tissues(ileum,cecum and colon) were removed for histological studies.Results:Both early and delayed-onset diarrhea can be observed on rats,the incidence of diarrhea in herb treated group were 1.83±0.72 compared with diarrhea group 2.19±0.94.The change of body weight and food-intake in herb group were better than diarrhea group(P<0.05,P<0.01).The histological changes were milder.Conclusion:Zingiberis Recentis Decoction for Purging Stomach-Fire can prevent the delayed-onset diarrhea caused by CPT-11.更多还原
