葛根异黄酮类成分对缺氧复氧损伤心肌细胞保护作用的研究

【摘要】 目的建立葛根异黄酮类成分的HPLC指纹图谱,分析其对心肌细胞缺氧复氧损伤保护作用的相关性。方法HPLC-TOF-MS法对葛根异黄酮各指纹峰进行辨认,并考察不同剂量葛根异黄酮给药组对乳鼠心肌细胞缺氧复氧损伤的保护作用,然后采用灰色关联分析法将峰面积和细胞存活率药效数据相关联。结果从HPLC指纹图谱中鉴定出7个葛根异黄酮类成分,分别为葛根素、3’-羟基葛根素、葛根素木糖苷、3’-甲氧基葛根素、葛根素-7-木糖苷、大豆苷和大豆苷元,这些成分的药效并无很大差异,相关系数最大值葛根素(0.761 4)与最小值大豆苷(0.670 8)之间相差不到0.1。结论葛根异黄酮保护缺氧复氧损伤心肌细胞的作用可能是通过多组分协同而发挥疗效。

【Abstract】 AIM To establish the HPLC fingerprint of pueraria isoflavones and to analyze the relationship between these constituents and their protective effect on hypoxia / reoxygenation injury myocardial cells. METHODS HPLC-TOF-MS method was used to identify the fingerprint of pueraria isoflavones,and rat myocardial cells with hypoxia / reoxygenation injury were treated with different dosages of these constituents to research their protective effects. The grey correlation analysis was applied to associating the fingerprint peak areas with cell survival rates.RESULTS Seven peaks from HPLC fingerprint of pueraria isoflavones were found and elucidated as puerarin,3’-hydroxy puerarin,puerarin-xyloside,3’-methoxypuerarin,puerarin-7- wood glycoside,daidzin and isoflavoues aglycone. There was little difference in cell survival rate between the maximum( 0. 761 4,puerarin) and minimum value( 0. 670 8,daidzin). CONCLUSION The protective effect of pueraria isoflavones on hypoxia / reoxygenation injury myocardial cells may be due to the synergistic action of multi-constituents.
