黄精多糖对免疫抑制小鼠免疫功能影响的实验研究

【摘要】 研究黄精多糖(PP)对环磷酰胺(Cy)所致免疫抑制小鼠免疫功能的调节作用。将40只小鼠随机分为5组:正常、模型、PP低、中、高(100,400,800 mg/kg)剂量组。每日灌胃给药饲养20 d,期间于d 3,d 6,除正常组外,其它各组按6 mg/kg腹腔注射Cy。测定器官与体液相关免疫指标。结果表明:与正常组比,模型组造模成功。与模型组相比,PP给药组能显著修复免疫抑制小鼠的DTH反应、脏器指数、吞噬指数、溶血素指数(P<0.05),且呈明显的剂量依赖性;高剂量PP给药组小鼠血清中SOD活性得到显著恢复(231.71±7.75)U/mL、MDA含量显著降低(6.20±0.65)nmol/mL,到达或超过了正常组小鼠相应指标。因此,PP能有效逆转和修复由Cy所致的免疫抑制小鼠的免疫功能,可开发为肿瘤放化疗的辅助治疗剂。

【Abstract】 To investigate the effects of polygonatum polysaccharide(PP) on the immunological activities in the immunosuppressed mice,forty ICR mice were divided into five groups: normal control,model(immunosuppressive mice),and L-,M-,H-dosage(100,400,800 mg/kg) PP.The mice were given once per day for 20 days by intragastric administration of PP solution.Beyond normal group,the mice of other groups were injected through peritoneal cavity 6 mg/kg of cyclophosphamide(Cy) on 3rd,6th.The immunity indexes were determined as follows: the swelling of ears,phagocytic index of peritoneal macrophages,the hemolysin,the weight of immune organs,the SOD activity and the MDA content in serum.To compare with normal group,the model group of immunosuppressive mice were induced successfully by 6 mg/kg of Cy.To compare with model group,PP could repair and enhance significantly the response of delayed-type hypersensitivity,the organ index,Phagocytic index and hemolysin indexsof mice(P < 0.05),and there was an obviously dose-dependent relationship.Specifically,the activity of SOD serum(231.71 ± 7.75) U/mL in h-PP group almost reached or even exceeded that of normal mice.Additionally,contents of MDA(6.20 ± 0.65) nmol/mL were far lower than those of normal mice.The PP can reverse and repair obviously the immune function of the immunosuppressive mice induced by Cy.The polygonatum can be developed into the adjuvant therapeutic agent in chemotherapy.
