基于生物信息学分析TGFBI基因在头颈部鳞癌中的表达及其预后意义
Analysis of expression of TGFBI gene in head and neck squamous cell carcinoma and its prognostic significance based on bioinformatics
【摘要】 目的 探讨转化生长因子β诱导基因(transforming growth factor β-induced gene, TGFBI)在头颈部鳞癌(head and neck squamous carcinoma, HNSC)中的表达及其预后意义。方法 首先通过TIMER2.0数据库分析TGFBI基因在泛癌中的表达水平;并进一步利用UALCAN数据库和GEPIA数据库检索TGFBI基因在HNSC与正常组织中的表达水平,以及TGFBI基因与患者种族、年龄、性别、肿瘤分期、淋巴结转移状况等相关临床特征的关系;通过STRING数据库分析TGFBI基因的互作蛋白,借助DAVID数据库对其与互作蛋白基因进行功能富集分析;最后通过GEPIA数据库分析TGFBI基因在HNSC中的生存意义。结果 发现TGFBI在HNSC中的表达明显高于正常组织(P<0.05),TGFBI高表达的患者总生存期明显低于低表达的患者(P<0.05)。蛋白互作分析发现,FN1、MMP14、ITGA3、ITGAM、ITGAV、ITGB1、ITGB2、ITGB3、TGIF2和TGIF2LX这10个蛋白基因与TGFBI存在紧密联系,此外,TGFBI及相关基因功能富集分析显示它们主要参与了5个生物学程序、4个细胞组分、3个分子功能、4个信号通路。结论 TGFBI在HNSC中的表达水平较正常组织明显升高,且与患者预后不良有关,有望成为HNSC诊断和治疗的生物学靶点。
【Abstract】 Objective To investigate the expression of transforming growth factor β-induced(TGFBI) gene in head and neck squamous carcinoma(HNSC) and its prognostic significance. Methods First, the expression level of TGFBI gene in pan-cancers was analyzed by TIMER2.0 database. Furthermore, UALCAN database and GEPIA database were used to search the expression levels of TGFBI gene in HNSC and normal tissues and the relationship of TGFBI gene with relative clinical characteristics such as race, age, gender, tumor stage and lymph node metastasis status. The interacting proteins of TGFBI gene were analyzed by STRING database, and the functional enrichment analysis was performed on TGFBI gene and its interacting protein genes through DAVID database. Finally, the survival significance of TGFBI gene in HNSC was analyzed through GEPIA database. Results It was found that the expression level of TGFBI in HNSC was significantly higher than that in normal tissues(P<0.05), and the overall survival time of patients with high expression level of TGFBI was significantly lower than that of patients with low expression(P<0.05). Protein interaction analysis showed that there was a close relationship between TGFBI and 10 proteins: FN1, MMP14, ITGA3, ITGAM, ITGAV, ITGB1, ITGB2, ITGB3, TGIF2, and TGIF2LX. In addition, the functional enrichment analysis of TGFBI and related genes showed that they were mainly involved in 5 biological processes, 4 cell components, 3 molecular functions and 4 signal pathways. Conclusion The expression level of TGFBI in HNSC is significantly higher than that in normal tissues and is related to the poor prognosis of patients, which is expected to become a biological target for the diagnosis and treatment of HNSC.
