乳酸杆菌后生元体外抗诺如病毒感染的作用及机制初探
Against norovirus infection effects and preliminary mechanisms of action of postbiotics derived from probiotic Lactobacillus in vitro
【摘要】 目的 探讨乳酸杆菌所制备的后生元是否具有体外抑制鼠诺如病毒(mouse norovirus, MNV)感染细胞的作用,并进一步初步探究其通过干扰素(Interferon, IFN)途径抑制病毒感染的作用机制。方法 培养三株鼠李糖乳杆菌、一株格氏乳杆菌分别制备后生元制剂,即无菌培养上清液和菌体裂解液,通过CCK8方法检测后生元干预下MNV感染的鼠源巨噬细胞Raw264.7的细胞活性动态变化,RT-qPCR技术测定鼠李糖乳杆菌所制备的后生元与MNV共作用于Raw264.7细胞时,细胞IFN-β和IFN-λ表达水平随时间的变化。结果 后生元对病毒有显著抑制作用,菌体裂解液对病毒的抑制效果优于培养上清液。IFN-β基因相对表达量在培养24 h时最高,IFN-λ基因相对表达量较少且在48 h时达到峰值。结论 体外实验表明,乳酸杆菌所制备的后生元可在体外显著抑制MNV复制。我们推测后生元,主要是裂解后菌体成分,是通过刺激宿主细胞IFN-β的表达从而达到抗MNV感染的效果,下一步需要对具体抗毒成分及干扰素途径调控机制进行研究。
【Abstract】 Objective To investigate whether postbiotics derived from probiotic Lactobacillus can inhibit murine norovirus(MNV) infection in vitro. Moreover, to further explore the preliminary mechanisms of action of postbiotics anti-virus whereby interferon(IFN). Methods We used Cell Counting Kit-8(CCK8) to detect the interaction with cell culture supernatant, cell lysate, and MNV on Raw264.7 cells, with the bacteria of three strains of Lactobacillus rhamnosus and one strain of Lactobacillus gasseri. The expression of IFN-β and IFN-λ in a strain of Lactobacillus rhamnosus was determined by RT-qPCR when cultured with MNV-1 in RAW264.7. Results Postbiotics showed a significant inhibition of MNV. The inhibitory effect of cell lysates on norovirus was better than that of cultured supernatants. The relative expression of IFN-β reached its peak at 24 h, while the relative expression of IFN-λ reached its peak at 48 h. Conclusion In vitro experiments with Raw264.7 cells showed that, the postbiotics could significantly inhibit MNV infection, and the specific inhibitory components are located in the lysed bacteria. We hypothesize that postbiotics, especially the bacteria lysate, could achieve anti-MNV effect by stimulating the expression of IFN-β in host cells, which is worth exploring the specific antivirus components and the regulatory mechanism of interferon pathway in the next step.
