外源激素对荷花生长及相关基因表达的影响

【摘要】 为了探讨植物激素对荷花生长发育的影响,以荷花‘青玉’为试验材料,分析了在赤霉素GA3(90、120、150mg/L)和细胞分裂素6-BA(100、150、200mg/L)处理后,荷花生长发育状态、酶活性、丙二醛含量和相关基因表达量的情况。结果表明:1)150mg/L GA3和100mg/L 6-BA处理使荷花叶片的叶面积的增加效果最为显著,150mg/L GA3和150mg/L 6-BA处理使荷花株高的增长量最大。2)GA3和6-BA处理均有利于提高荷花花瓣的酶活性和丙二醛含量,浓度120mg/L GA3和150mg/L 6-BA处理下作用最佳。3)EXP基因的表达水平呈现先上升后下降的趋势,在盛花期达到峰值。而CKX基因表达水平逐渐增加,由盛花期至谢花期表达量迅速提升,达到最大值。表明120mg/L GA3和150mg/L 6-BA能有效促进荷花的生长发育,起到显著延长花期的作用。

【Abstract】 In order to understand the effects of exogenous hormones on the growth and development of lotus,a lotus cultivar‘Qingyu’was treated by gibberellin(GA3)with the concentrations of 90,120,and 150 mg/L,and cytokinin(6-BA)with concentrations of 100,150,and 200 mg/L.Relative parameters were measured,such as the state of plant development,enzyme activity,malondialdehyde content and related gene expression levels.The results showed that:1)the treatments of 150 mg/L GA3 and 100 mg/L 6-BA had the most significant effect on the increase of leaf area.The treatments of 150 mg/L GA3 and 150 mg/L6-BA resulted in the largest increase of height.2)Both GA3 and 6-BA treatments were beneficial to the increase of the enzyme activity and malondialdehyde content of lotus petal,and the effects were best under the treatments of 120 mg/L GA3 and 150 mg/L 6-BA.3)The expression level of EXPgene increased first,reached the peak value at the full-flowering stage and then decreased afterwards.The expression level of CKXgene gradually increased and rose rapidly from full-flowering stage to flower-wilting stage,reaching the maximum value in full-flowering stage.These results suggested that 120 mg/L GA3 and 150 mg/L 6-BA treatments could effectively promote the growth and development of lotus,and played significant roles in prolonging the flowering period.
