彗星实验评价氯乙烯作业工人职业性DNA损伤的危险因素
Risk factors assessment on occupational DNA damage in vinyl chloride workers using comet assay
【摘要】 目的分析氯乙烯作业工人彗星实验结果,评价氯乙烯作业工人DNA损伤主要影响因素。方法分离外周血淋巴细胞,经彗星实验分析DNA损伤情况,应用聚合酶链反应(PCR)检测谷胱甘肽转移酶(GSTT1,GSTM1)基因型;应用PCR和限制性酶切片段长度多态分析技术(RFLP)检测细胞色素P4502E1酶基因型(CYP2E1),收集历年体检资料,然后对结果进行统计分析。结果经各因素同OTM做秩和检验,平均每年接触量、CYP2E1c1c2/c2c2、饮酒均与氯乙烯作业工人DNA损伤有关。结论氯乙烯工人DNA损伤情况受遗传特性、生活习惯、氯乙烯作业等因素的综合影响,故用于氯乙烯工人健康监护应作全面考量。
【Abstract】 Objective Investigating the comet assay results of VCM (vinyl chloride monomer) workers to evaluate the main influencing factors of DNA damage by this method. Method Comet assay was used to detect the DNA damage.The genotypes of GSTT1,GSTM1and CYP2E1were determined by polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP) respectively in DNA of white blood cells in the workers.Result Rank test showed that CYP2E1c1c2/c2c2,average VC exposure level and alcohol-drinking habit were all correlated with DNA damage.Conclusion DNA damage was affected by lifestyle,genetic character and VCM exposure.Therefore,the finding of DNA damage should be given comprehensive analysis in occupational health surveilliance of vinyl chloride workers.
