四逆汤配方颗粒和传统饮片汤剂的有效成分比较

【摘要】 目的 比较四逆汤配方颗粒及其传统汤剂中甘草苷、甘草酸、6-姜酚、6-姜烯酚、乌头碱、新乌头碱的含量。方法 采用HPLC法检测四逆汤配方颗粒及其传统饮片汤剂中甘草酸、甘草苷、6-姜酚、6-姜烯酚、乌头碱及新乌头碱的含量,分析主要成分在两者中的含量差异,采用C18色谱柱,流动相为乙腈-0.05%磷酸水,梯度洗脱,柱温30℃,检测波长237 nm,流速1.0 mL·min-1,进样量10μL。结果 四逆汤中甘草苷、甘草酸、新乌头碱、乌头碱、6-姜酚及6-姜烯酚等成分均在测定范围内线性关系良好(r分别为0.9994、0.9997、0.9991、0.9993、0.9992、0.9993),四逆汤传统片汤剂中甘草苷、甘草酸、新乌头碱、乌头碱、6-姜酚、6-姜烯酚的平均回收率分别为98.16%(RSD=1.0%)、100.6%(RSD=l.9%)、98.58%(RSD=1.4%)、98.28%(RSD=1.3%)、100.5%(RSD=1.4%)、99.01%(RSD=1.9%);四逆汤配方颗粒中各成分的平均回收率依次为99.15%(RSD=1.3%)、98.81%(RSD=1.1%)、100.4%(RSD=1.4%)、98.87%(RSD=1.2%)、98.14%(RSD=1.8%)、99.28%(RSD=1.4%)。四逆汤传统汤剂中甘草苷、甘草酸、新乌头碱、乌头碱、6-姜酚、6-姜烯酚质量分数分别为1.627~1.637、1.057~1.075、0.671~0.682、0.540~0.550、0.659~0.678、0.0251~0.0256 mg·g-1;配方颗粒中6种成分含量分别为1.866~1.910、2.765~2.826、1.177~1.210、0.989~0.994、0.368~0.373、0.0461~0.0472 mg·g-1。结论 与传统汤剂比较,四逆汤配方颗粒中甘草苷、甘草酸、6-姜烯酚、新乌头碱及乌头碱的含量较高,6-姜酚的含量较低。四逆汤配方颗粒较好地保留了饮片中绝大多数的指标成分,为配方颗粒在临床上代替传统饮片使用提供了依据。

【Abstract】 OBJECTIVE To contrast the contents of liquiritin, glycyrrhizic acid, mesaconitine, aconitine, 6-gingerol, 6-shogaol and the pharmacodynamic differences in formula granule and traditional decoction of Sini decoction.METHODS The contents of liquiritin, glycyrrhizic acid, mesaconitine, aconitine, 6-gingerol, 6-shogaol in the extracts obtained from two preparation methods of Sini decoction were determined by HPLC on a C18 column.The mobile phase consisted of acetonitrile-0.05% phosphoric acid in water with gradient elution.Column temperature was set 30 ℃ with a detection wavelength of 237 nm, flow rate of 1.0 mL·min-1,and injection volume of 10 μL.RESULTS The components of liquiritin, glycyrrhizic acid, mesaconitine, aconitine, 6-gingerol, 6-shogaol in Sini decoction were all linearly within the range of determination(r were 0.9994,0.9997,0.9991,0.9993,0.9992 and 0.9993).The average recoveries of liquiritin, glycyrrhizic acid, mesaconitine, aconitine, 6-gingerol, 6-shogaol in traditional decoction of Sini decoction were 98.16%(RSD=1.0%),100.6%(RSD=l.9%),100.5%(RSD=1.4%),99.01%(RSD=1.9%),98.58%(RSD=1.4%) and 98.28%(RSD=1.3%);The average recoveries of each ingredient in formula granules of Sini decoction were 99.15%(RSD=1.3%),98.81%(RSD=1.1%),100.4%(RSD=1.4%),98.87%(RSD=1.2%),98.14%(RSD=1.8%) and 99.28%(RSD=1.4%).The contents of the six constituents in Sini decoction traditional decoction were 1.627-1.637,1.057-1.075,0.671-0.682,0.540-0.550,0.659-0.678,0.0251-0.0256 mg·g-1,respectively.And in the formula granule of Sini decoction were 1.866-1.910,2.765-2.826,1.177-1.210,0.989-0.994,0.368-0.373,0.0461-0.0472 mg·g-1,respectively.CONCLUSION Compared to the traditional decoction, the contents of liquiritin, glycyrrhizic acid, mesaconitine, aconitine and 6-shogaol in formula granule of Sini decoction had greater levels, but the content of 6-gingerol remained lower.The formula granules of Sini decoction retained most of the index components in decoction pieces, which provided experimental basis for the formula granules to replace traditional decoction in clinical use.
