黄芪总提取物促进PBMC对肝癌细胞的体外杀伤作用

【摘要】 目的观察黄芪总提取物(TAE)与人外周血单个核细胞(PBMC)联用对肝癌细胞株BEl-7402的体外杀伤作用。方法 MTT法及流式细胞术分别检测TAE与PBMC在单独、联合应用情况下对BEL-7402细胞的杀伤活性,Burgi修正公式评价药物联合应用对细胞杀伤效果是否具有协同作用,ELISA法检测TAE对PBMC分泌IFN-γ、TNF-α细胞因子的影响。结果 TAE与PBMC联用对肝癌细胞BEl-7402具有协同杀伤作用。TAE预先与PBMC孵育后两者协同杀伤作用明显增强。TAE可以刺激PBMC分泌IFN-γ、TNF-α细胞因子。结论 TAE可以促进PBMC对肝癌细胞的体外杀伤能力,二者联用具有协同杀伤作用,其作用机制可能是TAE刺激PBMC分泌IFN-γ和TNF-α以增强免疫杀伤能力。

【Abstract】 Objective To investigate the synergistic cyctoxic effect of total astragalus extract( TAE)combining with human peripheral blood mononuclear cells( PBMC) on BEL-7402 cells in vitro. Methods MTT assay and flow cytometry were employed to test the cytotoxicity of TAE with or without PBMC on BEL-7402 cells. The modified Burgi formula was used to analyze the effect of two treatments. ELISA was used to detect the IFN-γ and TNF-α level of PBMC after TAE treatment. Results TAE combining with PBMC produced a synergistic cytotoxic effect on BEL-7402 cells. The synergistic degree was increased when PBMC was pretreated with TAE. TAE induced the secretion of IFN-γ and TNF-α by PBMC. Conclusion TAE enhances the cytotoxicity of PBMC against liver cancer cells. TAE combining with PBMC may produce a synergistic cyctotoxic effect,by which mechanism is related to upregulation of IFN-γ and TNF-α secretion and cytotoxicity of PBMC.
