生物发酵技术在再造烟叶降糖中的控制和应用
Control and Application of Biological Fermentation Technology to Reduce Sugar Contents in Reconstituted Tobacco
【摘要】 为实现再造烟叶总糖含量的可调可控,以再造烟叶生产线提取液为研究对象,利用高通量测序技术对再造烟叶提取液进行测序分析,获得了提取液内真菌和细菌的种群分布以及优势菌株,然后以优势菌株酿酒酵母为菌种,考察了不同发酵条件对提取液总糖含量的影响,同时探索了膜过滤技术在终止发酵反应中的应用。结果表明:酿酒酵母属为再造烟叶浓缩液真菌种群中的优势菌属,乳酸杆菌属为细菌种群中的优势菌属;以酿酒酵母为菌种,在一定温度和时间下,浓缩液总糖含量随着酵母添加比例的上升而逐渐下降;100 nm陶瓷膜过滤添加酿酒酵母后的提取液,过膜后提取液经过24 h存放,总糖含量维持稳定,说明膜过滤方式能够终止发酵反应。
【Abstract】 In order to control and adjust the total sugar content in reconstituted tobacco, the extraction liquid from production line was studied, and Illumina MiSeq sequencing platform was used to analyze 18 S rDNA sequences in fungi and 16 S r DNA sequences in bacteria. The community composition of fungi and bacteria, together with the dominant strains were obtained. Then, the effect of different fermentation conditions on the total soluble sugar content of extraction liquid was investigated with saccharomyces cerevisiae as the dominant strain. At the same time, the application of membrane filtration technology in stopping fermentation reaction was explored. The results showed that the dominant genera of fungi included saccharomyces cerevisiae. The dominant genera of bacteria included Lactobacillus. At a certain temperature and time, Saccharomyces cerevisiae was used as the strain, the total sugar content reduced with the increase of the proportion of added saccharomyces cerevisiae. 100 nm ceramic membrane was used to filtrate extract liquid which adding saccharomyces cerevisiae, then went through 24 h storage, the total sugar content remained unchanged.
