基于网络药理学探讨附子理中汤治疗非酒精性脂肪性肝病作用机制

【摘要】 目的:运用网络药理学方法分析附子理中汤(FZLZD)治疗非酒精性脂肪肝性病(NAFLD)的潜在作用机制。方法:检索TCMSP数据库筛选出FZLZD药物活性成分及作用靶点,并借助Uniprot平台进行靶点蛋白规范化处理。检索GeneCards与OMIM数据库得到NAFLD靶点基因,进一步获取药物和疾病共有靶点,即为FZLZD治疗NAFLD的潜在靶点基因。利用Cytoscape 3.7.2绘制药物活性成分-靶点-疾病网络图,利用Cytoscape 3.7.2结合String数据库构建蛋白互作(PPI)网络,利用Metascape数据库对共有靶点进行GO和KEGG富集分析。结果:最终筛选出80种药物活性成分、43个潜在作用靶点;富集分析获取808个生物学过程及25条与NAFLD相关信号通路。结论:FZLZD通过多成分-多靶点-多信号通路协同调控NAFLD。

【Abstract】 Objective:To explore the mechanism of treatment of non-alcoholic fatty liver disease(NAFLD) with Fuzi Lizhong Decoction(FZLZD).Methods:The active components and action targets of FZLZD were screened by searching the TCMSP database, and the proteins were standardized by the Uniprot database. The targets of NAFLD, obtained by searching GeneCards and OMIM databases, were taken intersection analysis with drug action targets to get the potential targets of FZLZD in the treatment of NAFLD. Then, Cytoscape 3.7.2 was used to conduct the network of active component-target-disease, and the protein-protein interaction(PPI) network was constructed through the String database and Cytoscape software to infer the core target. GO annotation analysis and KEGG pathway analysis was conducted through the Metascape bioinformatics annotation database. Results:In this study, there were 80 main traditional Chinese medicine active components of FZLZD and 43 potential targets for NAFLD treated by FZLZD; FZLZD in the treatment of NAFLD involved 808 GO and 25 KEGG pathways. Conclusion:FZLZD played an important role in treatment of NAFLD, through multiple components, multiple targets, and multiple pathways.
