不同处理对四川牡丹种子萌发的影响
Effects of Different Treatments on Seed Germination of Paeonia decomposita
【摘要】 通过探究不同基质和不同浓度的GA3处理对四川牡丹种子萌发的影响,揭示四川牡丹种子萌发的最佳处理条件,旨在为四川牡丹种群扩繁和保护提供科学依据。本研究在生根阶段测定了不同处理条件(基质:细沙、椰糠、草炭土∶蛭石∶珍珠岩=1∶1∶1;GA3浓度:0、50、100、150、200 mg/L)下四川牡丹的生根参数;在发芽阶段测定分析了不同根长区间(L1:主根长度≤60 mm;L2:主根长度> 60 mm)、不同基质(细沙、细沙∶黄壤=1∶1、草炭土∶蛭石∶珍珠岩=1∶1∶1)、不同浓度(0、50、100、150 mg/L)的GA3条件下四川牡丹的发芽参数。结果表明:1)生根阶段,细沙和GA3处理能使四川牡丹提前生根,提高生根率。细沙和200 mg/LG A3组合,四川牡丹生根历期最短(21d),生根率最高(1000%)。2)发芽阶段,根长区间、GA3浓度、根长区间和基质的交互作用对四川牡丹发芽率有极显著影响;四川牡丹种子发芽的优化条件为:选用主根长度> 60 mm的种子,置于细沙∶黄壤=1∶1土壤中,用50mg/L GA3处理,发芽率最高(933%),发芽速率最大(516 d-1)。细沙和GA3处理对四川牡丹生根有促进效应。不同根长区间、基质和GA3浓度(发芽)处理可有效打破四川牡丹上胚轴休眠,缩短初萌期,提高发芽率,促进种子发芽。
【Abstract】 By investigating the effects of different substrates and GA3 concentrations on seed germinationofPaeonia decomposita, the best condition for seed germination was revealed in order to provide scientificbasis for the population expansion and protection ofP. decomposita. The rooting parameters ofP. decompositawere determinedunder different treatment conditions(substrates: fine sand, coconut bran, peat soil∶ vermiculite∶ perlite = 1∶ 1∶ 1; GA3 concentration: 0, 50, 100, 150, 200 mg/L) during the rootingstages; in the germination stage, the germination parameters ofP. decompositawere determined and ana-lyzed under different root length intervals(L1: main root length≤ 60 mm; L2: main root length > 60 mm), different substrates(fine sand, fine sand∶ yellow soil = 1∶ 1, peat soil∶ vermiculite∶ perlite = 1∶ 1∶ 1) and different GA3 concentrations(0, 50, 100, 150 mg/L). The result showed: 1)In rooting stage,fine sand and GA3 treatment could makeP. decompositarooting earlier and it could increase the rootingpercentage ofP. decomposita. With the combination of fine sand and 200 mg/LGA3,P. decomposita rooting period was the shortest(21 d), the rooting percentage was the highest(100. 0%). 2)During thegermination stage, interaction of root length interval and substrate, the root length interval and GA3 con-centration had significant effects on the germination percentage ofP. decomposita. The optimum condi-tions for germination ofP. decompositawere as follows: seeds with main root length > 60 mm were selected and placed in the substrate(fine sand∶ yellow soil = 1∶ 1 soil), and treated with GA3 at 50 mg/L concentration. By this method, the highest germination percentage(93. 3%) and the highest germina-tion rate(5. 16 d-1) of P. decompositawere obtained. Fine sand and GA3 treatment had a promotingeffect on the rooting ofP. decomposita. Different root length interval, substrate and GA3 concentration(germination) treatment could effectively break the dormancy ofP. decompositaepicotyl, shorten the ini-tial germination period, increase germination percentage and promote seed germination.
