肺泡巨噬细胞对豚鼠气道平滑肌神经兴奋的影响

【摘要】 以调理酵母多糖(OPZ)激活的大鼠肺泡巨噬细胞(AM)上清液,作用于受电场刺激的豚鼠单个气管环平滑肌标本。结果显示,对气道平滑肌(ASM)胆碱能神经兴奋有易化作用,其机理可能是AM释放前列腺素类物质(PGs)促进了接头前膜ACh的释放。AM上清液对肾上腺素能和非肾上腺素能抑制神经活动没有明显影响,但使ASM舒张恢复50%时程(RT1/2)缩短,提示在病理情况下AM大量激活时,可导致气道口径缩小、阻力增大;这可能在哮喘的发病中起一定作用。

【Abstract】 Alveolar macrophages(AM) were isolated from Wistar rats by lung lavage and activated byopsonized zymosan(OPZ).The supernatant of their culture was collected.Trachea smoothmuscle(TSM) preparation from a single ring of trachea was placed in a 12 ml organ bath con-taining Krebs solution equilibrated with 95 % O2 and 5% CO2,and the maximal shortening(△Lmax)elicited by electric field stimulation (EFS)in the presence of propranolol(10-6mol/ L)and indomethacin(10-5mol / L)or the maximal relaxation(-△Lmax)in the presence of atropine(3×10-6mol / L)were measured. It was found that the OPZ activated AM supernatant increasedthe contraction elicited by the cholinergic nerve excitation and this enhancement could be abol-ished by indomethacin-treated AM supernatant. AM supernatant showed no effect on the con-eentration-dependent response of TSM to exogenous acetylcholine(ACh). The relaxation elic-ited by adrenergic and nonadrenergic inhibitory nerve excitation was not significantly influencedby AM supernatant, but the recovery duration of the maximal relaxation was shortened. It wassuggested that the enhancement of contraction by AM supernatant may be mediated byprostaglandins(PGs).PGs released by AM may facilitate the prejunctional cholinergicneurotransmission on airway smooth muscle,and contribute to the pathogenesis of bronchialasthma.
