表面活性剂协同超声波提取泡桐花总黄酮及其抗氧化活性研究

【摘要】 采用表面活性剂十二烷基硫酸钠(SDS)协同超声波法提取泡桐花总黄酮,在单因素实验的基础上,采用响应面法优化泡桐花总黄酮的提取工艺,并评价其抗氧化活性。结果表明,泡桐花总黄酮的最佳提取工艺为:SDS质量分数0.55%、提取温度60℃、提取时间65 min、超声功率240 W,在此条件下,泡桐花总黄酮提取率为64.98 mg·g-1,与预测值(65.03 mg·g-1)接近;泡桐花总黄酮具有较强的DPPH自由基、ABTS自由基清除能力及铁还原力,对DPPH自由基、ABTS自由基的IC50值分别为0.07 mg·mL-1、0.10 mg·mL-1

【Abstract】 We extracted total flavonoids from Paulownia flowers by surfactant(sodium dodecyl sulfate, SDS)-ultrasonic method.Moreover, we optimized the extraction process of total flavonoids from Paulownia flowers by response surface methodologies on the basis of single-factor experiments, and evaluated its antioxidant activity.The results show that the optimal extraction conditions can be determined as follows: the SDS mass fraction of 0.55%,the extraction temperature of 60 ℃,the extraction time of 65 min, and the ultrasonic power of 240 W.Under above conditions, the extraction rate of total flavonoids from Paulownia flowers can reach 64.98 mg·g-1,which is close to the predicted value(65.03 mg·g-1).The total flavonoids from Paulownia flowers has strong scavenging ability to DPPH free radicals and ABTS free radicals and iron reducing ability, and the IC50 values of total flavonoids from Paulownia flowers towards DPPH free radicals and ABTS free radicals are 0.07 mg·mL-1 and 0.10 mg·mL-1,respectively.
